Is it possible to create a one-day only course in REM?

Yes but it requires some configuration. Go to Course Offerings List settings and modify columns for the Access Period Start and End Dates:

 

in the Additional Column Settings > Date and Time Format change 'Date Only' to 'Date & Time'.
